Ok, new kid on the block. I have what may be a dumb question, but I can't seem to find an answer anywhere. Maybe it's because it's more obvious that I think. I think I understand this, but I want to check with the experts first.
From what I understand the BuckPuck controls leds. But so does the PC. Except the PC also controls sound. That's the only big difference, right? So if I use a PC instead, I won't need a BuckPuck?

You are correct. The PC has a LED driver built into the board. You will not need a BuckPuck with the PC.

Could you use a Buck Puck with the Nano Biscotti instead of a resistor for the main LED or is that overkill ?

No. You need the resistor. They're smaller, cheaper, etc, etc, etc.

Not to mention that the Buckpuck needs more than 5v to operate, and the NB's maximum voltage rating is 5.5v. You'll either underpower the buckpuck, or cook the NB.
